So I want to purchase the 12-hole 330cc injectors and I'm wondering if they are plug-and-play or do you have to recalibrate the ECM to accommodate the higher fuel flow?

Are you SCed? I thought on the site it only reccomended the 250-270cc for 06.
I just did mine, after you have the battery disconnected for a time period the ecu will reset and need to relearn parameters and driving habbits(shift points) but you will also experience misfires due to lack of fuel/pressure when you first restart the truck.

What I would do if I was to do it again, disco the battery, do the injectors, at the same time clean the MAF and throttle body(a tech told me this is important, especially when the computer it trying to relearn) then connect the battery and only cycle the key to on to build fuel pressure; I had to drive mine 10 miles or so before I felt I had proper fuel pressure, the whole time untill then you could definitely feel it missing.
I would then clear any codes that might of come about from misfires, and then disconnect the battery again so it relearns with no codes and proper fuel pressure.
    She drives great now!

Quick answer: Shifting points seem more precise. No searching delay on downshift/deceleration. Doesn't make sense because the transmission is separate. Maybe has to do with ECM readings, but not sure exactly.

Reason I originally did this was an exercise in experimentation. I wanted to clean my existing injectors, but that was a 3 day minimum turn around time. Buying brand new injectors was crazy expensive. So, I tried these remans after talking with Keith who has a 2002 as well IIRC. The cost of remans was very fair and equal with just pro injector cleaning. I had the knowledge base from him so gave it a go. Took about 5 hours. Do not do these for MPG gain. Its more for a modern shifting point feel.

Those 330cc injectors are for if you only run straight ethanol iirc and I don't believe they are plug n play. However the 250cc for non vvti and 270cc for vvti are plug n play
