Hello! I have a few questions: (1) How do I know if my 07 Tundra SR5 is a 1st or 2nd Gen? and (2) My transmission overfill valve will not turn when pressure is applied.

Definitely 2nd Gen then. You have the 5.7 and the newer body style. Best to ask any questions you have on that forum. Bievenido.
