My 2006 crew cab, the door lock buttons don't do anything. If I lock a door by hand then open a door with the key in the ignition it does unlock the doors.
So I don't think it would be a fuse, but I have looked at the owners manual and checked all the ones I thought might have something to do with them.

ETA: After a little more messing, the window switches on the driver's door don't work either. This is my wife's truck I don't drive it often.
So I'm guessing there are some broken wires somewhere.

    Any suggestions?
    Thanks

There's contacts in the switches that can wear over time as well. I've read of some replacing the switches. Could be as simple as a loose connector. Needs some diagnostic inquiry performed on it. Pull the switch and inspect.

Time to take the panel off and poke it with the multimeter.
