I don't think there's any adjustment. My guess would be that it needs to be lubed. Remove the interior door skin and use white lithium grease (or something similar) to spray any pivot points or joints. The most likely problems would be at the door handle, and at the latch.

The "inside the door" components have no adjustments. The only place the door can be adjusted is where the lock meets the striker on the B-Pillar. All that usually does is adjust the fit of the door to the body. It is worth trying if the fit is loose (door rattle?) when the door is closed. Just loosen teh two striker bolts and move it in towards the truck center a bit with light hammer blows. If the door is fitting well to the body, don't adjust this now.

The plastic bushings where the lock rods go between the handle and the door lock are likely getting sloppy from wear. I wish I had some pics to help explain, but I don't own a Tundra. However, I know the structure pretty well. They are small plastic pieces that hold the rods to the handle and the lock. See the "Snap" pieces in the link. Your handle may also be worn, but hard to say without taking it apart.

Doors come apart pretty easy. Pull the switch base out (window up first), remove a couple screws (handle and armrest), pull the trim panel loose and remove the handle from the panel leaving the cables attached. These rest of the parts are a little hard to access. Play around with it and see where the slop is before taking it all the way apart.

One other thing, you didn't explain if it's hard to open from inside or outside. I am assuming outside with this advice. If from inside, that same type snap can get worn on the inside handle. You'll see it if you take the door apart. Both locations, you rotate them to unlock them, then the rod slips out. Pinch the back side and push and they'll pop out.

    Good luck.
