Rear main seal. Mine leaks to. Worth a look at the valve covers since they are often leaky and easier to replace.

I concur. Looks like it could be a rear main seal to me.

The bell housing appears damp pretty high up, which would be less suspicious for rear main seal, maybe you'll get lucky and it's just valve covers.

Is this a hole? Is it supposed to be there?

Remove the inspection plate and see if it’s clean behind there.

I have a leak like that, it stains the same part but is coming from the oil cooler o-rings (to the right of the oil filter). The leak runs along the top of the oil pan seal and hits the same spot as yours. Waiting to fix on my next oil change. I loose about half a quart of oil every 4K miles. Iv had this leak for about 20k miles now. I couldn’t find where it was coming from

Are you loosening any fluids, is it painting/staining the floor? Is it oil or coolant or both

That’s not a horribly difficult fix and a cheap rubber seal to buy OEM. The Toyota Maintenance channel on YouTube has a video dedicated to it. Sorry if you know this already.

Yeah, I watched the video so many times now. Just gearing up the courage to tackle the job. On the 06 there seams to be a hidden bolt behind the ac compressor that’s not fun to get to. Also trying to line up the repair with an oil change.
