Hello, i jacked up my rear end with both wheels off of the ground. I put the truck in drive. The rear right wheel is making a noise. It sounds slightly like a growl noise. My question is, should the tires be making any noise at all with it off the ground like that? There is also play in both of my rear tires.

Growling noise usually indicates a bad bearing, so I suspect you have a bad wheel bearing or two. The tires should have nothing to do with the growling noise, while safely jacked up take the wheels off to verify whether or not the noise is still there. You can put a lug nut or 2 on the studs to make sure the drums stay in place.
When you say you have play in both tires, do you mean play in rotation or play like 12 and 6 o'clock in and out? There should be no play if checking it like you would the front ball joints (12 and 6 o'clock).

There is 3 to 9 o clock play and 12 to 6 o clock play. I have a super noticeable noise when driving too. Its most prominent at 47 mph and only happens when I'm on the throttle. Its a pulsating deep growling/ low pitched hum. Will a wheel bearing make noises at certain speeds?

Yeah, that type of play concerns me and sure sounds like a bad wheel bearing. When I've had bad wheel bearings they were on front wheels and they got louder as the speed increased. When I had a bad pinion bearing on my Ford van the sound was only really noticeable while decelerating.

Just seems like waaayyyy to big of a coincidence to happen right after doing my carrier bearing.. @Professional Hand Model was the noise more prominent at certain speeds?
