I was wondering if I need an alignment and if I need my speedometer adjusted after I install a leveling kit?

Alignment yes. Speedometer only if you went with bigger tires and even then, not a big deal if you don’t.

weirdly, my speedo went 3 mph off after lowering the truck, same stock wheels/tires. Would consistently read 45 on speedo, when radar signs clocked me at 42mph

    Too many, but not enough....
your stock speedo is probably actually off by +2 mph directly from the factory, so actually going up to 33" tires from the stock 32" size will bring it closer to true speed.

You sound like a bot...? It's a certainty that it will effect alignment... on a Tundra by a lot.
