2001 Tundra 2wd V6. Speedo/odo not working. Replaced both speed sensors at AT trans and now replaced combination meter (entire cluster). Still nothing! Can anyone help please?!

Did you get a CEL? PO500 by chance? Here is a test for your VSS first:

Check this website, its for Tacoma but I'm pretty sure its identical for testing purposes to your Tundra.
    http://118.69.35.147/VR-Tools/TSKT/...al/Diagnostics/Circuit Inspection/cip0500.pdf

Key take away, is the next step is to check the SP1 @ back of ECU to see if you're getting the signal from there. I know you said your speedo isn't working but checking to see whether or not that pin has the proper voltage (signal to ECU) will verify/back-up what you're (not) seeing on the combo meter. Sometimes, just sometimes, these steps are necessary because you can actually have a bad connection/harness/pin on the plug going to the cluster.

The only thing I'm not sure of on your Tundra is whether or not the VSS signal first goes through the ABS comp or direct to the combo meter. Do you know the answer to that one? If it goes direct to the meter and you know how to detect shorts with your multimeter than you may be able to find something there on the way to the metro from your VSS.

Is your ABS performing normally? Did you pull your original cluster apart by chance and find anything unusual like cracked solder? Harness/wiring from VSS @ tranny looks good?

According to everything I’ve read, the VSS with the gear sends its 4-pulse signal directly to the combination meter, and the other VSS, which appears to be electro magnetic, sends its signal to the ECU.
As far as I can tell, my base model truck has no ABS sensors.
I visually inspected the cluster and saw no issues. I can’t tell what wires to test at the back of cluster, as there appear to be no less than 6 wires with the same color combo as those at the VSS.

Download this FSM: https://drive.google.com/file/d/0B9w76cywnmguanBiSEtCb0RkaGM/view and pass on the Karma to someone else sometime.

You'll have to look into the BE (body electrical) section and find the diagram for your setup to determine where to look next on how the signal is created and where it goes. If your sensors are good than you'll need to follow the next location so on and so forth. It looks like the pin you're looking for is C13 - the C plug is the 2nd from the left when looking at the back of the cluster. If you unplug that connector from the cluster and you're looking at the portion that makes contact with the cluster, all the way on the right is PIN #13 - that should be your VSS input.

BE-48 through B-53 will be a good place to start. Those will be all the pages you need to correctly diagnose your issue.
